脐橙与柠檬种间细胞电融合再生杂种植株

SOMATIC HYBRID PLANT REGENERATION BETWEEN NAVEL ORANGE AND LEMON VIA PROTOPLAST ELECTRO FUSION

  • 摘要: ‘纽荷尔’脐橙(Citrussinensis (L.) Osbeck)胚性细胞悬浮系原生质体与‘尤力克’柠檬(Citruslim on (L.) Burm.f.)叶肉原生质体经电场诱导融合,培养8 个月,首批再生了1棵植株。形态学观察、染色体计数及RAPD分析证明为异源四倍体体细胞杂种。该杂种的获得为三倍体无籽柠檬品种培育提供了杂交亲本。

     

    Abstract: Protoplasts isolated from cell suspension cultures of ‘Newhall' navel orange (Citrus sinensis (L.) Osbeck) were electrically fused with mesophyll protoplasts of ‘Eureka' lemon (Citrus limon (L.) Burm.f.).One plantlet was regenerated after 8 months of culture.Morphological observation,chromosome counting and RAPD analysis confirmed that this plant was allotetraploid somatic hybrid.This somatic hybrid will be utilized as possible pollen parent for breeding of seedless triploid lemon cultivars.
