栗种间杂交系的同工酶基因与形态标记的遗传连锁关系研究

LINKAGE RELATIONSHIPS OF ISOZYME GENES AND MORPHOLOGICAL MARKERS IN INTERSPECIFIC CROSSES OF CHESTNUT

  • 摘要: 探讨了美州栗(Castaneadentata Borkh)与中国栗(C.mollissimaBlume)种间杂交的1个F2代和2个回交一代家系中8个同工酶基因:Acp3、Est1、Est5、Prx1、Prx2、Prx3、Me和Adh与2个形态标记:Inh和Twh的连锁遗传关系。研究结果表明:在所有家系中, Inh与Prx1和Est5具有一致的连锁关系, 其基因的连锁顺序为Inh-Prx1-Est5.还发现在参试的3个家系中有1个家系的4个基因对(Acp3-InhAcp3-Prx1、Me-InhTwh-Inh)是连锁的, 因此, 有2个同工酶基因和2个形态标记初步定位成1个连锁组, 其基因顺序为Twh-Inh-Prx1-Est5.证明在多年生木本果树上, 同工酶基因可以通过连锁分析与形态基因整合为1个单一基因图而无需另外的杂交。还讨论了在用不同的栗杂交家系寻找连锁遗传关系时结果不一致等问题。

     

    Abstract: Linkage relations among eight isozyme genes, Acp3, Est1, Est5, Prx1, Prx2, Prx3, Me and Adh, and two morphological markers, Inh and Twh, were investigated in one F2 and two BC1 families of interspecific crosses between the American chestnut (Castanea dentata Borkh)and the Chinese chestnut (C.mollissima Blume), Inh was found to be consistently linked with Prx1 and Est5 in all families. The order of these three genes was determined to be Inh-Prx1-Est5.In addition, four other gene pairs, Acp3-Inh, Acp3-Prx1, Me-Inh and Twh-Inh were found to be linked in one of the three families Investigated.The two isozyme genes and two morphological marker genes were tentatively integrated into one linkage group with the following gene order Twh-Inh-Prx1-Est5.This study demonstrated that isozyme genes can be integrated with morphological marker genes into a single linkage map without the need for additional crosses.the inconsistencies observed in detecting linkage relationships among the chestnut families were discussed.
