EMBRYOGENESIS AND ORGANOGENESIS ON CULTURED ANTHERS OF HYOSCYAMUS NIGER

  • 摘要: 天仙子花粉在附加不同浓度 NOA 的 BN 培养基中,形态发生能力不同,低浓度易由花粉直接形成胚状体,高浓度一般先形成愈伤组织,再由愈伤组织分化胚状体或根芽。实验表明,直接起源于花粉的胚状体与经花粉愈伤组织形成的胚状体,其胚胎发生的模式和顺序相同,均由原胚、球形胚、心形胚、鱼雷期胚到子叶期胚,但二者原胚的起源不同。花粉愈伤组织亦可经器官发生途径产生根芽。

     

    Abstract: The ability of morphogenesis in anther culture of Hyoscyamus niger was quite different when anthers were cultured in Bourgin and Nitsch's(BN)medium supplemented with fifteen gradient concentrations(0.01=100mg/L)of β-naphthoxyacetic acid(NOA). Three kinds of developmental pathway were observed: 1. pollen grain gave rise to embryoids directly; 2. pollen grain initiated callus from which embryoids developed and 3. buds and roots were produced from pollen callus.It was cvident from the results that the pattern of development was similar to cither embryoids derived from pollen grain directly or from callus. The embryoids were differentiated through typical globular-shaped, heartshaped; torpedo-shaped and cotyledonary stages similar to those of zygotic embryos. The differences between two kinds of embryogenesis are the origin of proembryoid.The initiation of bud and root from pollen callus are discussed.
