南瓜属植物RAPD-PCR反应体系的建立与应用

Optimization and Application of RAPD-PCR System for Cucurbita

  • 摘要: 采用混合DNA样品池,利用正交法对南瓜属植物RAPD反应条件进行了探讨,并利用建立起来的优化体系,从35条引物中筛选出7条引物,对供试中国南瓜、印度南瓜和黑籽南瓜3个栽培种的7个品种进行了RAPD分析,结果均获得了较多的多态性位点。RAPD-PCR反应扩增结果还显示,日本南瓜兼有中国南瓜和印度南瓜的特征带,聚类分析的结果则表明,它与中国南瓜的亲缘关系更近。

     

    Abstract: Using DNA pooling,we established RAPD optimal conditions for Cucurbita through the orthogonal optimization method.With the founded system,7 primers were selected form 35 primers to analyze 7 cultivars of C.moschata Duch,C.maxima Duch and C.ficifolia Bouche.The results showed that there were many polymorphic sites among the cultivars.The cultivar Riben Nangua had the distinct bands of C.moschata and C.maxima at the same time,and the results of cluster analysis showed it was near with C.moschata.
