植物生长调节剂对荷叶铁线蕨不同器官离体繁育的影响

Effects of plant growth regulators on in vitro propagation of different organs of Adiantum nelumboides X. C. Zhang

  • 摘要: 以荷叶铁线蕨(Adiantum nelumboides X. C. Zhang)走茎茎尖、无菌原叶体及孢子体为实验材料,分析植物生长调节剂对不同器官材料离体繁育的影响。结果显示,激动素(KT)与吲哚丁酸(IBA)可以促进走茎茎尖的正常生长和发育,6-苄氨基嘌呤(6-BA)可以促进生长点的增殖,而2,4-二氯苯氧乙酸(2,4-D)则促进生长点分化形成愈伤组织;0.5 mg/L的6-BA在促进原叶体形成绿色圆球茎(Green globular body,GGB)及GGB的增殖方面效果最佳,1.0 mg/L的玉米素(ZT)在诱导GGB分化方面表现最优;0.5 mg/L的萘乙酸(NAA)处理下孢子体生根率最高;液体MS培养基中添加浓度为0.5 mg/L的NAA有利于GGB的成苗分化和孢子体的生长。本研究探究了不同植物生长调节剂对荷叶铁线蕨不同器官离体繁育生长发育的调控作用,为规模化繁育荷叶铁线蕨提供了实践指导。

     

    Abstract: This study evaluated the regulatory effects of plant growth regulators on in vitro propagation of distinct organ types in Adiantum nelumboides X. C. Zhang, using rhizome tips, sterile gametophytes, and sporophytes as explants. Results showed that kinetin (KT) and indole-3-butyric acid (IBA) promoted normal shoot tip growth, while 6-benzylaminopurine (6-BA) most effectively promoted shoot meristem proliferation. Application of 2,4-dichlorophenoxyacetic acid (2,4-D) induced callus formation from growing points. In gametophyte cultures, 0.5 mg/L 6-BA maximized the induction and proliferation of globular green bodies (GGB), whereas 1.0 mg/L zeatin (ZT) most effectively promoted GGB differentiation. In sporophyte regeneration, 0.5 mg/L naphthaleneacetic acid (NAA) yielded the highest rooting rate and enhanced both GGB-derived seedling differentiation and sporophyte growth in liquid MS medium. These findings clarify the organ-specific hormonal responses in A. nelumboides and offer a foundational framework for optimizing large-scale ex situ propagation protocols for this endangered species.
